"hayatimdan birşey(ler) eksik" is good. something -> birşey or birşeyler depending on your thinking style. 1 thing or more.

hayatımda eksik birşey(ler) var. --> Is a non-logical but correct sentence as it is in English. There is --> var.

If something is missing how can it be there (is) ?

that is how we say it in english, even though it may sound non-logical to say "there is something missing" in your opinion when concerning turkish rules, it is gramatically correct
