In Türkçe, how would one say:
I am looking very forward to hearing back from you and I hope you have a lovely weekend.
(using "siz" and "sen")
And how do you end a letter/email in Turkish? Would it just be a translation of sincerely? Or something else?

You can end a letter ´saygılarımla´ with respect or respectfully.

"Cevabını sabırsızlıkla bekliyorum. Umarım güzel bi hafta sonu geçirirsin."
you can end with "Saygılarımla" "respectfully" and for formal end.
if your friends "Sevgilerimle" would be better.

Cevabını dört gözle bekliyorum ve umarım güzel bir hafta sonu geçirirsiniz.
PS 1: If he/she is your friend you can use "cevabını", but if he/she is your boss, or someone in a higher rank; or if you write to more than one person you could use "cevabınızı". This is the same for "geçirisiniz", if he/she is one person it should be "geçirirsin".
PS 2: At the end, you can use "Sevgilerimle, or sevgiler"(Love), or "Saygılarımla, or saygılar"(Sincerely)

By the way, "dört gözle beklemek" does not mean "waiting with four eyes"  It´s an idiom and means "look forward to".
