it means he/she knows that you don't want to
istediği/n/i means that you want where -n is possesive suffix for 2nd person singular and -i is acusative, which means there should be a verb like bilmek, which requires acusative suffix

bilir - s/he knows
istememek - not to want

to say "s/he knows you don't (didn't) want" you have to use a THAT clause.

To construct it, you take the verb's root - isteme - in this case and add a -dik- participle followed by possessive ending (-in) and direct object ending (-i):

isteme-dik -in-i

and, finally, you have to make use of the consonant harmony (k becomes ğ if followed by a vowel)

and then you have

isteme-diğ-in-i bilir

any clearer now?
