No,actually you cann't use yok here.
Yok means not exist,so you cann't use it instead of Hayır,which means No.

So when you want to say something in İstek Kipi,you use same suffixe as Emir Kipi with sen,O,Siz,Onlar ?
Doğru mu ?

When you want to say,
Let him got to school,
We say,
Okula gitsin.
Not,
Okula gele.

Doğru mu ?
